Tryptic Soy Broth (TSB), also known as Soybean-Casein Digest (SCD) Medium, is a general nutrient and enrichment broth. It is used in qualitative procedures for the cultivation of fastidious and non-fastidious microorganisms and is also recommended for sterility testing. TSB is recommended for use as the inoculum broth for disc diffusion and agar dilution antimicrobial susceptibility testing by the CLSI. Middlebrook 7H11 Agar is a selective and non-differential medium used for the isolation and cultivation of mycobacteria. Cohn et al. modified Middlebrook 7H10 by adding enzymatic digest of casein and determined organism growth was stimulated for fastidious strains of Mycobacterium tuberculosis. The use of agar-based media for primary isolation of mycobacteria have two significant advantages. (1) They do not usually liquefy in the presence of contaminating proteolytic organisms. (2) They retain exact concentrations of added drugs because the medium is solidified with agar rather than by inspissation of the egg. Bile Esculin Azide Agar with Vancomycin and Streptomycin (BEAVS) is a selective and differential medium used for isolation and presumptive identification of group D streptococci, Enterococcus faecalis, and Enterococcus faecium. Group D streptococci and enterococci hydrolyze esculin in the presence of bile, resulting in esculetin and dextrose. Esculetin reacts with ferric ions to form brown-black colonies. Oxgall (bile salts) inhibit most strains of non-group D streptococci and enterococci. The medium is supplemented with 8µg/mL Vancomycin and 100µg/mL Streptomycin. Streptomycin inhibits gram-negative organisms present in background flora. Blue-white screening is a rapid and efficient technique for the identification of recombinant bacteria.
